I will be using the Uzumaki clan as reference to my theory. The Uzumaki clan and the Senju clan are relatives. Both of them are related in an unknown way. The Uzumaki clan had formed an alliance with the Senju clan.
Uzushiogakure was continuously growing in power and became one of the most feared villages. The other nations grew wary of their power. Their power became the reason for their downfall.
It was completely destroyed by the other shinobi nations. The few people who were alive left the village. As we know that Hashirama was a very kind man, who was always looking forward to help others. So, it’s really simple that he would help his precious allies.
So, during this battle, Hashirama fought alongside the Uzumaki clan. His power would be a big threat to any army of the world. Hashirama must’ve fought against the armies of the other nations.
But, as we know that the other nations had the advantage of numbers, it would’ve been difficult. The countries would have surely sent their strongest warriors to fight for them. Going against several Jōnin level shinobi would be difficult, even for Hashirama Senju. This is where I think Hashirama was killed.
